Cellulase Enzyme Production By Aspergillus Niger And Trichoderma Reesei On Different Lignocellulosic Wastes

Wheat straw, rice straw and rice husk were used as lignocellulosic substrates for the production of cellulase enzyme using Aspergillus niger and Trichoderma reesei. From fermentation studies, rice straw gave the best resultswith a CMCase activity of 0.61IUml-1 for T.reesei and 0.60IUml-1 for A.niger, FPUof 0.42IUml-1 for T.reesei and 0.50IUml-1 for A.niger compared to wheat straw(0.47IUml-1 CMCase and 0.32IUml-1 FPU for T.reesei and 0.46IUml-1 CMCase and 0.45IUml-1 FPU for A.niger) and in rice husk(0.49IUml-1 CMCase and 0.38IUml-1 FPU for T.reesei and 0.40IUml-1 CMCaseand 0.30IUml-1 FPUfor A.niger).Optimumconditions like pHand temperature for enzyme fromeach fungi were studied, 6 pH and 300C temperature for T.reesei and 5.5 pH and 320C for A.niger were found to be optimum. Molecular weight of these enzymes were determined by SDS PAGE and it was found to be 30 kDa for T.reesei and 40 kDa for A.niger.
